A permeapad® assisted sample preparation workflow for the selective determination of anticonvulsants in human serum by liquid chromatography tandem mass spectrometry.

The aim of this project was to develop a green sample preparation workflow using Permeapad® 96-well plates for the determination of five anticonvulsants in human serum. The biomimetic membrane within the Permeapad® facilitated selective drug permeation from the complex serum matrix, significantly reducing organic solvent consumption. The subsequent LC-MS analysis demonstrated excellent linearity (R2 > 0.98) across a wide range of concentrations, allowed effective chromatographic analysis. Recovery studies confirmed selective permeation with aqueous donor conditions proving most effective. This Permeapad® approach presented an efficient and eco-friendly alternative to therapeutic drug monitoring, paving the way for high-throughput bioanalysis.
